He is the son of Virginia Roberta, a psychic, and William Raymond "Billy Ray" Thornton, an educator, high school history teacher and basketball coach. He has two younger brothers named Jimmy Don and John David.

Thornton's first success as an actor came when he co-wrote and starred in the 1992 film One False Move. His other notable films include Sling Blade (1996), Armageddon (1998), Monster's Ball (2001) and The Astronaut Farmer (2006). He has also appeared in several television series, including The Outsiders (1983) and Hearts Afire (1992–93).

In addition to acting, Thornton is also a musician and songwriter. He released his first album Private Radio in 2001 and has since released three more albums; Hobo (2003), The Edge of the World (2007) and Beautiful Door (2010). In 2007 he formed his own band called the Boxmasters with fellow musician Jody Stevens.

He has won several awards throughout his career including an Academy Award for Best Adapted Screenplay for Sling Blade in 1996, a Golden Globe Award for Best Actor – Motion Picture Musical or Comedy forPrimary Colors in 1998 and a Critics' Choice Movie Award for Best Actor in a Drama Film for Monster's Ball in 2001.
